85% RPMI 1640 + 15% FBS split semi-confluent (2/3 to 3/4 confluency) culture 1:10 every 7 days using trypsin/EDTA (for 10-20 min); seed out at ca. 1-2 x 106 cells/80 cm 2 , adherence may require several hours or overnight at 37C with 5% CO2 cell harvest of about 20 x 106 cells/175 cm 2;
doubling time of about 50-70 hours;
