DMEM + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Selected for 6-thioguanine and ouabain resistance; sensitive to HAT; HPRT negative. Used routinely as feeder layers prepared by irradiation or mitomycin-c treatment for maintenance of teratocarcinoma stem cells in the undifferentiated state.
