Part of the Business Division of the Health Protection Agency (ECACC, Salisbury, Wiltshire)
DMEM + 2mM Glutamine + 10-15% Fetal Bovine Serum (FBS). Pre-soak flasks in 0.1% gelatin for over 2 hours and wash with PBS. Split confluent cultures 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin EDTA; 5% CO2, 37C. Upon resuscitation cells shed a lot of debris, leave for 2 days before subculture. Cells will attach with or without the use of collagen.

Derived from testicular carcinoma. F9 undergo very limited differentiation under normal conditions but can be induced to differentiate into parietal endoderm in the presence of retinoic acid and dibutyryl cAMP.

PROPERTIES: The cells undergo very limited differentiation under normal culture conditions, but can be induced to differentiate into a) parietal endoderm in the presence of retinoic acid (RA) and dibutyryl cyclic AMP; b) visceral endoderm when cultured in suspension as aggregates and treated with RA. Differentiation markers are: tissue type plasminogen activator (tPA) for parietal endoderm cells; alpha fetoprotein (AFP) and urokinase plasminogen activator (uPA) for visceral endoderm cells. In addition, both cell types lose the stage specific embryonic antigen-1 (SSEA-1), and synthesize large amounts of type IV collagen, laminin and fibronectin.

passage 6 after in vivo passage; split every two days; leave the cells with trypsin for one or two minutes at room temperature; the flasks must be presoaked with 0.1% gelatin for 10 minutes
