Ham's F12 + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:3 to 1:6 i.e. seeding at 1-3x100,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Karyotype: 2n = 42
Derived from a rat glial tumour induced by N-nitrosomethylurea. The cells produce S-100 protein which is found in the brain and also in brain tumours from both human and in other animals. The cell line is capable of performing this organ specific function over a period of a year and is capable of producing elevated levels of glyceryl phosphate dehydrogenase activity when stimulated by glucocorticoides. The synthesis of S-100 is dependent on the level of confluence and increases ten fold as cells grow to confluence.
