Description: human neuroblastoma established from the supraorbital metastasis of a neuroblastoma of a 14-year-old girl in 1971 (see ATCC HTB 10); although described in the original paper as neuroblastoma, SK-N-MC is now widely regarded as having originated from the morphologically similar Askin's tumor related to Ewing's sarcoma; FLI1-EWS gene fusion, the molecular pathological change in Ewing's sarcoma and Askin's tumor, was shown in SK-N-MC by Dunn et al. (Cancer Genet. Cytogenet. 76: 19-22, 1994) confirmed as human with IEF of AST, MDH

90% Dulbecco's MEM + 10% FBS + 2 mM L-glutamine + 1x MEM non-essential amino acids split confluent culture 1:3 to 1:6 every 3-5 days using trypsin/EDTA; seed out at ca. 1-4 x 106 cells/80 cm 2 ; cells may grow slowly initially at 37C with 5-10% CO2 cell harvest of about 15 x 106 cells/175 cm 2;
doubling time of ca. 48 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
