SPECIES: human, Caucasian
TISSUE/ORGAN: cervix
TUMOR: carcinoma
DSMZ catalog code: ACC 136
Morphology: adherent cells, epithelial-like, growing as monolayers
Susceptibility to viruses: adenovirus 3 * herpes simplex * influenza * poliovirus 1 * vaccinia
Description: human cervix carcinoma (derivative of HELA) originally described as 'epidermoid carcinoma established from the mouth of a Caucasian man in 1954'; however, as shown by DSMZ DNA fingerprinting (see below) this cell line must be considered an HELA subclone (human cervix carcinoma, see HELA DSM ACC 57) confirmed as human with IEF of AST, MDH
Viruses: ELISA: reverse transcriptase negative; PCR: EBV-, HBV-, HCV-, HIV-, HTLV-I/II-
Depositor: Dr. J. Wehland, GBF, Braunschweig, Germany
General Restrictions
Available in the following LABORATORY:
-
German Collection of Microorganisms and Cell Cultures (DSMZ, Braunschweig)
90% MEM (with Earle's salts) + 10% FBS + MEM non-essential amino acids + 0.075% Na-bicarbonate split confluent culture 1:3 to 1:5 every 3-6 days using trypsin/EDTA at 37C with 5% CO2 about 5-10 x 106 cells/80 cm 2 flask;
frozen with 70% medium, 20% FBS, 10% DMSO at about 3 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (hypertriploid karyotype with 8% polyploidy; 76(73-78)<3n>) * DNA fingerprinting (same Hinf I-(gtg)5 DNA profile as HELA) * immunological analysis (cytokeratin+ (100%) desmin- endothel- GFAP- neurofilament- vimentin++ (100%)) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase negative)
