85-90% McCoy's 5A + 10-15% FBS split confluent culture 1 x per week at up to 1:5 using trypsin/EDTA; seed out directly in a 80 cm 2 culture flask, cells will reach confluency within 5-8 days; cells may clump after trypsinization and may be difficult to disperse at 37C with 5% CO2 cell harvest of about 5-10 x 106 cells/80 cm 2;
doubling time of about 36 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (human near diploid karyotype with tetraploid sideline (31%) and 3.5% polyploidy; 46(41-46)<2n>XX; no consistent chromosome abnormality detected) * DNA fingerprinting (unique DNA profile with (gtg)5 multilocus probe) * immunological analysis (cytokeratin- desmin- endothel- GFAP- neurofilament- vimentin+) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase negative)
