Collection of human fibroblasts and UV sensitive CHO mutants


Two collections are available: the first contains human fibroblast strains derived from skin biopsies, the second selected mutants derived from the hamster cell line CHOK1. All cell lines have been established in the course of research programs on DNA repair mechanisms in mammalian cells. Human fibroblasts (group 1) are from patients affected with inherited defects in DNA repair processes and/or cancer proneness and from their relatives. The majority are homozygous or heterozygous for a xeroderma pigmentosum mutation (XP complementation groups A, C and D). Hamster clones (group 2) are hypersensitive to DNA damaging agents, UV light in particular; they have been selected by conventional mutagenesis, enrichment in repair defective mutants, isolation of colonies with reduced capability to synthesize DNA or to grow after UV irradiation. Both CHO mutants and human fibroblasts were characterised for survival and UDS after exposure to UV lights or to chemical mutagens, and for the frequency of chromosome aberrations. Genetic analysis of the DNA repair defects was performed by complementation studies.
