DMEM + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:20 i.e. seeding at 1x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
FLYA13 has been developed together with FLYRD18 (95091902) for packaging of retrovirus, it is especially useful for in vivo gene therapy . Both cell lines have been derived from HT1080 cells (85111505). FLYA13 has been transfected with Moloney murine leukaemia virus cores with envelope glycoproteins of amphotropic murine leukaemia virus. This cell line is able to produce human complement-resistant retroviral vectors at titres consistently higher than 1x10,000,000 i.u./ml. Addition of 4µg/ml blasticidin S and 10µg/ml phleomycin into the media may enhance the packaging function. No replication competent virus has been detected in this cell line.
