Endogenous viruses: contains Epstein-Barr
Description: human Burkitt's lymphoma established from the Burkitt's lymphoma (left orbital biopsy) of a 16-year-old African boy in 1967; cells were described to be EBV-positive and to express mRNA for proto-oncogene bcl-2; this cell line has been assigned by the ZKBS (German Central Commission for Biological Safety) to risk category 2 confirmed as human with IEF of AST, NP, PEP B
Viruses: ELISA: reverse transcriptase negative; PCR: EBV+, HBV-, HCV-, HHV-8-, HIV-, HTLV-I/II-
Depositor: Dr. J. Minowada, Fujisaki Cell Center, Okayama, Japan

90% RPMI 1640 + 10% FBS maintain at 0.5-1.0 x 106 cells/ml; split ratio 1:2 to 1:4 every 2-4 days; cells may grow slowly initially; cell culture may be started in 24-well-plates and later transfered to flasks; seed out at ca. 0.5-1 x 106 cells/ml at 37C with 5% CO2 maximal density at about 1.7 x 106 cells/ml;
doubling time of about 30 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
contamination was eliminated with Mycoplasma Removal Agent, then negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (near diploid with 20% polyploidy; 46(45-48)<2n>XY/XXY, +7, -9, t(8;14)(q24;q32)) * DNA fingerprinting (unique Hinf I-(gtg)5 DNA profile) * immunological analysis (CD3- CD10+ CD13- CD19+ CD37+ HLA-DR+ smIgG- smIgM+ sm+ sm-) * isoenzymes (confirmed as human with IEF of AST, NP, PEP B) * reverse transcriptase (reverse transcriptase negative)
