90% RPMI 1640 + 10% FBS + 2 mM L-glutamine + non-essential amino acids + 1 mM sodium pyruvate + 9 ęg/ml bovine insulin maintain at 0.3-1.0 x 106 cells/ml; split ratio 1:3 to 1:5; avoid lipopolysaccharide contamination which will result in cell clumping, maturation and retarded growth; cells are best maintained in 24-well plates; seed out at ca. 0.3 x 106 cells/ml at 37C with 5% CO2 maximal density at ca. 1.0 x 106 cells/ml;
doubling time of ca. 60 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
