85-90% Dulbecco's MEM + 10-15% FBS split confluent culture 1:2 to 1:4 about once or twice per week using trypsin/EDTA; seed out at ca. 2 x 106 cells/75 cm 2 at 37C with 5-10% CO2 cell harvest of about 20 x 106 cells/175 cm 2;
doubling time of ca. 33-60 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1 x 106 cells/ampoule;
contamination was eliminated with BM-Cyclin (tiamulin & minocycline), then negative in DAPI, microbiological culture, RNA hybridization assays;

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Fetal Bovine Serum (FBS) or Ham's F10 + 10% Horse Serum (HS). Split confluent cultures 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Karyotype: Modal No. 48; hyperdiploid
Derived from the Morris Hepatoma but is distinct from HTC. Immunological experiments demonstrate that the cultures synthesise and secrete rat serum albumin and synthesise tyrosine aminotransferase. Functions of the cells include the conjugation and metabolism of testosterone and bilirubin and the production of serum complement component, especially C9.
