85% McCoy's 5A + 15% FBS split confluent culture 1:2 to 1:3 every 2-3 days using trypsin/EDTA; seed out at ca. 1-2 x 106 cells/80 cm 2 in 10 ml medium at 37 C with 5% CO2 cell harvest of about 10 x 106 cells/175 cm 2;
doubling time of about 43 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (human karyotype) * DNA fingerprinting (unique DNA profile with (gtg)5 multilocus probe) * immunological analysis (cytokeratin -, desmin -, endothel -, GFAP -, neurofilament -, vimentin + (100%)) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase negative)
