90% RPMI 1640 + 10% FBS (cells may also be grown, but less efficiently, with MEM) maximal split ratio of 1:4; cells grow partly adherent, but trypsin is not needed to detach the cells at 37C with 5% CO2 about 46 x 106 cells/175 cm 2;
doubling time of ca. 25 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (74(69-76); rodent marker chromosomes present) * DNA fingerprinting (unique Hinf I-(gtg)5 DNA profile) * isoenzymes (confirmed as rat with IEF of AST, NP)
