Description: rat kidney fibroblasts cell line was cloned from a mixed culture of normal rat kidney cells; cells exhibit contact inhibition and are very sensitive; indicator cells for viral or chemical transformation; this cell line is frequently used for TGF-ss bioassays; (= ATCC CRL 1570) confirmed as rat with IEF of AST, NP, PEP B

95% Dulbecco's MEM + 5% FBS split culture 1:4 to 1:5 about 2-3 times a week using trypsin/EDTA when the cells have reached 3/4 of confluence at 37C with 5% CO2 cell harvest of about 3.5 x 106 /80 cm 2;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (near-diploid rat karyotype with 4% polyploidy; 40(37-42)<2n>) * isoenzymes (confirmed as rat with IEF of AST, NP, PEP B)

Part of the Business Division of the Health Protection Agency (ECACC, Salisbury, Wiltshire)
DMEM + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 5-10% Fetal Bovine Serum (FBS) or New Born Calf Serum (NBCS). Split confluent cultures 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C. The cells should be maintained sub-confluent, otherwise they will transform.

Derived from normal kidney and exhibits contact-inhibition. It is sensitive to viral or chemical transformation, including proteins such as SGF. Originated from the same mixed culture as NRK-52E but has distinct characteristics.
