90% RPMI 1640 + 10% FBS maintain at 0.2-1.0 x 106 cells/ml; split 1:2 to 1:4 every 2-3 days; seed out at ca. 1 x 106 cells/ml at 37C with 5% CO2 maximum density at 1.0-1.5 x 106 cells/ml;
doubling time of ca. 38 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
contamination was eliminated with Ciprobay (ciprofloxacin), then negative in DAPI, microbiological culture, PCR assays;

IDMEM + 2mM Glutamine + 20% Fetal Bovine Serum (FBS). Maintain cultures between 3-9x100,000 cells/ml; 5% CO2; 37C.
Karyotype: 2n = 46, diploid
Derived from the bone marrow aspirate of a 59 year old male Caucasian with erythroleukemia that became acute myelogenous leukaemia. The cells form colonies in soft-agar in the presence of colony stimulating factor (CSF). KG-1 cells resemble acute myelogenous leukaemia showing considerable pleomorphism with a predominance of myeloblasts and promyelocytes. A small percentage of the cells are mature granulocytes with the occasional presence of macrophages and eosinophils. They do not express surface Ig or EBV antigens and lack specific markers for lymphocytic cells.
