80% RPMI 1640 + 20% FBS + 1x non-essential amino acids split confluent culture 1:2 every 3-5 days (trypsin should not be used, cells can be detached by tapping the flask or pipetting); seed out at ca. 0.5-1.0 x 106 cells/cm 2 at 37C with 5% CO2 about 1-2 x 106 cells/ml or 0.5 x 106 cells/cm 2;
doubling time of ca. 40-50 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (hyperdiploid karyotype; 49(44-51)<2n>XY, +1, +6, +12, der(1)t(1;2HSR)(p33/35;?p22)x2, der(16)t(2;16)(p21/22;q24/25); HSR regions probably represent amplification of the N-myc oncogene which maps to 2p22 involved in the t(2;16) als) * immunological analysis (cytokeratin- desmin- endothel- GFAP- neurofilament+ vimentin++) * isoenzymes (confirmed as human with IEF of AST, MDH, PEP B) * reverse transcriptase (reverse transcriptase negative)
