90% Dulbecco's MEM + 10% FBS split confluent culture at 1:3 every 3 days or at 1:6 every 5-6 days using trypsin/EDTA at 37C with 5% CO2 cell harvest of about 50-60 x 106 cells/175 cm 2; doubling time of about 25-30 hours; frozen with 70% medium, 20% FBS, 10% DMSO at about 2 x 106 cells/ampoule; negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (near diploid hamster karyotype with tetraploid sideline (18%) and 18% polyploidy; 44(40-45)<2n>X/XX; paucity of homologous chromosomes indicates a high level of rearrangement; hamster markers present) * DNA fingerprinting (unique DNA profile with (gtg)5-probe, heterozygous with human single locus probe pYNH24 probe) * isoenzymes (IEF of AST, LDH, MDH, PEP B could not confirm Syrian hamster as species, but ruled out human species) * reverse transcriptase (reverse transcriptase negative)
