90% Medium 199 (25 mM HEPES) with Earle's salts + 10% FBS + 2 mM L-glutamine split confluent culture 1:2 every 4-7 days using trypsin/EDTA in PBS without Ca 2+ /Mg 2+ at +4C for 5-10 min at room temperature (about 20C) without CO2 (keep lid of flask closed), cells can be stored temporarily (up to 1 week) at 4C about 5-15 x 106 cells/80 cm 2;
doubling time of about 40 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at 2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
