RPMI 1640 + 20% FBS + 2mM L-Glutamine + 10 ng/ml GM-CSF; Freezing medium: 90% FBS + 10% DMSO; Slit 1:2-1:3 every two days; 37C, 5% CO2, mycoplasma negative, HOECHST and PCR
The viability may be low after thawing and need about one week to recover. The cells grow also with Iscove's MDM medium instead of RPMI and with IL-3 (10 ng/ml) instead of GM-CSF.
Validated by: Multiplex PCR (tested against human, rat, mouse primers. Confirmed as human with cytochrome c oxidase subunit I (cox I) primers)
