Ham's F10 + 2mM Glutamine + 10% Tryptose Phosphate Broth + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:2 to 1:4 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37C; subculture at about 75% confluence; cells can be dislodged by tapping flask against the palm of hand; use of 0.5% polyvinylpyrrolidine (PVP) in trypsin solution can increase viability; if trypsin is used neutralize and centrifuge to remove additive; cell growth is very slow.
Derived from methylcholanthrene-induced fibrosarcoma of Japanese quail. The cells are susceptible to several viruses including sub group A avian sarcoma, avian encephalomyelitis, Marek's disease herpes virus and rabies virus.
