DMEM:F12 (1:1) + 2mM Glutamine + 50ng/ml Hydrocortisone + 5µg/ml Insulin + 5-10% New Born Calf Serum (NBCS). Split confluent cultures 1:10 to 1:20 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C; subculture twice weekly.
Karyotype: Polyploid 80-90 chromosomes, 8H mode
The rat mammary tumour cell line LA7 is a clonal derivative of Rama 25. The parent line was established from a dimethylbenz(a)anthracene- induced mammary cancer in Sprague-Dawley rats. LA7 produces spontaneous ridges in passages up to approximately 25. They have been described as intermediate cell type (F-precursor) between Rama 25 and fusiform cells. Used as lethally irradiated (6000 rads) feeder layer LA7 promote the proliferation and colony formation of primary and sub-cultured mouse mammary, mouse thymus and rat mammary cells. The cell line can be used as a feeder layer up to passage 80 at least.
