BME (EBSS) + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:5 to 1:20 i.e. seeding at 2x1,000 to 2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Derived by exposing C3H/10T1/2 clone 8 cells with 3-methylcholanthrene for 24 hours. Subsequently they lost the property of contact inhibition. Inoculation of 100,000 cells or more into syngenic mice has been reported to produce fibrosarcoma.
