90-95% Iscove's MDM + 5-10% FBS + 2 mM L-glutamine + 2.5 x 10-5 M mercaptoethanol split confluent culture 1:2 to 1:5 twice to three times weeky; cells are trypsin-sensitive, detach mechanically after loosening by incubating in PBS (without Ca 2+ or Mg 2+ ) for 10-15 min; seed out at ca. 0.2-0.5 x 105 cells/cm 2 at 37C with 5% CO2 maximum cell harvest of about 1 x 106 cells/cm 2;
doubling time of ca. 20-30 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1.0-1.5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, PCR assays;
