This is the complete description of the cell line that you selected.
By clicking on a generic term (e.g., SPECIES) you will go to the corresponding index (e.g., index of species), while by clicking on a specific term (e.g., retinoblastoma) you will go to the list of corresponding cell lines (e.g., retionoblastoma cell lines).
SPECIES: human, Caucasian
DSMZ catalog code: ACC 203
Morphology: adherent, fibroblast-like cells growing as monolayers
Description: human neuroblastoma established from the supraorbital metastasis of a neuroblastoma of a 14-year-old girl in 1971 (see ATCC HTB 10); although described in the original paper as neuroblastoma, SK-N-MC is now widely regarded as having originated from the morphologically similar Askin's tumor related to Ewing's sarcoma; FLI1-EWS gene fusion, the molecular pathological change in Ewing's sarcoma and Askin's tumor, was shown in SK-N-MC by Dunn et al. (Cancer Genet. Cytogenet. 76: 19-22, 1994) confirmed as human with IEF of AST, MDH
Viruses: ELISA: reverse transcriptase negative; PCR: EBV-, HBV-, HCV-, HHV-8-, HIV-, HTLV-I/II-
Depositor: Dr. H. G. Drexler, DSMZ, Braunschweig, Germany
Available in the following LABORATORY:
German Collection of Microorganisms and Cell Cultures (DSMZ, Braunschweig)
90% Dulbecco's MEM + 10% FBS + 2 mM L-glutamine + 1x MEM non-essential amino acids split confluent culture 1:3 to 1:6 every 3-5 days using trypsin/EDTA; seed out at ca. 1-4 x 106 cells/80 cm 2 ; cells may grow slowly initially at 37C with 5-10% CO2 cell harvest of about 15 x 106 cells/175 cm 2;
doubling time of ca. 48 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (pseudodiploid karyotype with 6% polyploidy; 46(40-47)<2n>XX, +8, -16, t(1;4) (p36;p13), t(2;3)(q24;q29), der(3)t(3;3)(p13;q22), del(10)(q25), t(11;22)(q24;q12) t(15;22)(q21;q12), add(17)(p12), add(21)(p12); dmin in 9% of cells;) * cytogenetics 2 (resembles published karyotype; carries masked 11;22 translocation recurrent in Ewing's and Askin's tumor.) * DNA fingerprinting (unique DNA profile with (gtg)5 multilocus probe) * immunological analysis (cytokeratin+ (5-10%), desmin-, endothel-, GFAP-, neurofilament-, vimentin+ (100%)) * isoenzymes (confirmed as human with IEF of AST, MDH) * reverse transcriptase (reverse transcriptase negative)
