DMEM:Ham's F12 (1:1) + 2mM glutamine + 1mM NaP + 20U/l insulin + 10% FBS Split confluent cultures 1:2 i.e. seeding at 4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37C. Media rapidly becomes acidic and should be changed every 3 days, although cells need to be subcultured only every 7-10 days.
Derived from an ascitic fluid sample taken from a 74 year old female with serous papillary adenocarcinoma in post chemotherapy relapse. At laparotomy disease present in peritoneum with involvement of greater omentum and appendix but not ovaries.
