85% RPMI 1640 + 15% FBS split confluent culture 1:4 every 5-6 days using trypsin/EDTA (occasional loosely adherent clumps can be detached by tapping the flask); seed out at ca. 2 x 106 cells/80 cm 2 at 37C with 5% CO2 cell harvest of about 10 x 106 cells/80 cm 2;
doubling time of about 60 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 2-4 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
Validated by: cytogenetics 1 (flat-moded, near-triploid karyotype; 69(63- 72)<3n>XXY/XXYY,+1,-4,-8,+9,+12,+15,-17,+18,-22, der(9)t(9;11)(p13;q11)t(11;22)(q24;q12)x2, add(14)(p11); carries two masked copies of 11;22 translocation associated with sarcoma of Ewing) * DNA fingerprinting (unique DNA profile with multilocus probe (gtg)5) * immunological analysis (cytokeratin-, desmin-, endothel-, GFAP-, neurofilament-, vimentin+) * isoenzymes (confirmed as human with IEF of AST, MDH, NP) * reverse transcriptase (reverse transcriptase not detected)
