DMEM + 4mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:3 to 1:5 i.e. seeding at 2-4x10,000 cells/ml using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C. Clumped cells have tendency to float.
SK-N-F1 has been obtained from a bone marrow metastasis of a male patient with neuroblastoma. The poorly differentiated and slow growing cells secrete different neuroblastoma specific peptides and synthesise choline acetyltransferase. Reactivity of anti-leukaemia-associated and anti-foetal brain-associated monoclonal antibodies could be shown.
