90% RPMI 1640 + 10% FBS cells are difficult to culture! maintain at high density (>2 x 106 cells/ml); cells grow slowly; minimum density at ca. 0.5 x 106 cells/ml; seed out at ca. 1 x 106 cells/ml with initially 20% FBS at 37C with 5% CO2 maximum density at ca. 3-5 x 106 cells/ml or higher;
doubling time of ca. 70 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 5 x 106 cells/ampoule;
contamination was eliminated with Ciprobay (ciprofloxacin), then negative in DAPI, microbiological culture, RNA hybridization, PCR assays;
