90% RPMI 1640 + 10% FBS split confluent culture 1:8 every 5-6 days using trypsin/EDTA; seed out with 1-2 x 106 /25 cm 2 flask; initially after thawing, cells may grow slowly and loosely adherent; at 37C with 5% CO2 cell harvest of about 0.3 x 106 cells/cm 2;
doubling time of about 50 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule;
contamination was eliminated with Ciprobay (ciprofloxacin), then negative in DAPI, microbiological culture, RNA hybridization assays;
