Ham's F10 + 2mM Glutamine + 15% Horse Serum (HS) + 2.5% Fetal Bovine Serum (FBS). Allow clusters of cells to settle, remove supernatant leaving 5-10ml of growth medium in a 25cm² tissue culture flask, to cover aggregate. Gently remove clusters and divide equally in to 2 25cm² flasks and add fresh medium to each; 5% CO2; 37C Cells do not form monolayers, but tend to grow in small floating clusters and are subcultured as intact aggregates.
Derived after alternate passage of LAF1 mouse pituitary tumour cells as tumours in animals and in cell culture. Cells produce adrenocorticotropic hormone (ACTH) at approximately the rate of 1000mU/mg of cell protein per week.
