RPMI 1640 + 2mM Glutamine + 10-20% Fetal Bovine Serum (FBS). Maintain cultures between 1-5x100,000 cells/ml; 5% CO2; 37C; at low density or they may differentiate. Freeze in 10% glycerol and 90% FBS. After 6 weeks in culture cells may differentiate.
Karyotype: Modal No 46, pseudodiploid
Peripheral blood lymphocytes obtained by lymphophoresis from 36-yr old female. 10% spontaneously differentiate, proportion enhanced by polar-planar compounds e.g. DMSO, butyrate, hypoxanthine, TPA, actinomycin D, Retinoic acid.
