RPMI 1640 + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Maintain cultures between 1-3x100,000 cells/ml; 5% CO2; 33C.
A temperature sensitive mutant isolated from the mouse mammary cell carcinoma cell line, FM3A (ECACC No. 87100804). Cells will Not double at 39C (Non-permissive temperature) and will grow at 33C (permissive temperature). Electron Microscope findings have shown that chromatin abnormally condenses into fragmented and compact forms in 80% of the cells at 39C for 16 hours. This abnormal condensation does Not occur at 33C or in the FM3A (ECACC No. 87100804) cell line at either temperature.
