85% RPMI 1640 or MEM (with Earle's salts) + 15% FBS split confluent culture every 2-3 days using trypsin/EDTA; for the test cells should be starved and split once a week; seed out about 1 x 106 cells/175 cm 2 at 37C with 5% CO2 cell harvest of about 15 x 106 cells/175 cm 2;
frozen with 70% medium, 20% FBS, 10% DMSO at about 2.5 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (apparently bovine karyotype with bimodal ploidy distribution; 42(40-42)<2n>/80(77-84)<4n>; structurally rearranged with apparent reduction of chr number by centric fusion followed by tetraploidization) * isoenzymes (confirmed as bovine with IEF of G6PD, NP)
