Description: mouse macrophages established from the bone marrow of a C3H/HeJ mouse confirmed as mouse with IEF of AST, LDH, NP, PEP B

80% RPMI 1640 + 20% horse serum + 1 mM sodium pyruvate + 1 mM non-essential amino acids + 1.25 æg/ml vitamin B12 + 75 æM alpha-thioglycerin + 2 mM L-glutamine + 2.5% 10x L-929 supernatant (DSM ACC 2) split 1:10 every 4-6 days detaching the cells by flushing the adherent cells with medium through a 25G needle (do not try to desaggregate cells by passing through the needle); cells grow best in Corning round plates (100 x 20 mm) at 37C with 5% CO2 cell harvest of about 3-5 x 106 cells/plate;
doubling time of about 50 hours;
frozen with 70% medium, 20% FBS, 10% DMSO at about 0.5-1.0 x 106 cells/ampoule;
negative in DAPI, microbiological culture, RNA hybridization assays;
Validated by: cytogenetics 1 (murine near-diploid karyotype with 10% tetraploidy; 42(37- 43)<2n>; centric fusion markers absent) * isoenzymes (confirmed as mouse with IEF of AST, LDH, NP, PEP B) * reverse transcriptase (reverse transcriptase not detected)
